Sample Prep:
Starting material (Eukaryotic): High Quality total RNA (5.0µg – 40.0µg)
cDNA synthesis protocol: Affymetrix recommends starting with a minimum of 5 µg of total RNA at a minimum concentration of 0.5 µg/µL.
Determining RNA Concentration: After purification the RNA concentration is determined by absorbance at 260nm on a spectrophotometer (one absorbance unit = 40 µg/mL RNA) The A260/A280 ratio should be approximately 2.0, with ranges between 1.8 to 2.1 considered acceptable.
